Yes. In recent years, To Kill a Mockingbird and the most recent revival of The Cherry Orchard featured musicians performing the score live.

I don't know about union rules.

A play that has a prominent music component (as opposed to more of a background score) often has live musicians, I think. One Man, Two Guvnors, Lady Day at Emerson's Bar and Grill and Farinelli and the King had live musicians, to cite recent examples.
