One big one for me is Doris Day as Mrs. Robinson in THE GRADUATE.
Anne Bancroft is magnificent, but the role WAS offered to Doris, and her husband talked her out of it.
When the subject of what might have been casting comes up, for some reason this one always leaps to mind.
